Mark Wahlberg will star as Nathan Drake in Uncharted: Drake's Fortune
"David is one of the best writer/directors I've ever worked with. The idea that he has is just insane. So hopefully we'll be making that movie this summer."
There was also speculation, first raised by Showbiz 411, that Robert De Niro and Joe Pesci were being eyed for roles in the movie, which Mark Wahlberg confirmed. He also revealed what roles they would play:
"I'm obviously in whatever David wants to do but the idea of it is so off the charts: De Niro being my father, Pesci being my uncle. It's not going to be the watered-down version, that's for sure."
Robert De Niro and Joe Pesci have not been confirmed to star in the film, but talks are ongoing. We'll be sure to keep you posted with any further information on Uncharted: Drake's Fortune as soon as we have it.
Uncharted: Drake's Fortune comes to theaters in 2012 and stars Mark Wahlberg, Robert De Niro, Joe Pesci. The film is directed by David O. Russell.

I think Walhberg is a good choice for the role. As much as Fillion would have been the best choice, the simple fact is that he was never going to get it because he doesn't have any box-office appeal outside of the fan community. In casting Wahlberg, they can market this as a broad Indiana Jones-style adventure film rather than as a video game adaptation.

Mark looks the part but I don't see him pulling of the smartassness & charm of Drake. I would've like to have seen either Chris Evans, Chris Pine, David Boreanaz(watch Bones), or Nathan Fillion get the role of Drake. Besided as much as I like De Niro & Pesci, keep them the hell out of this movie & don't add characters that weren't in the game. They would just seem soo out of place in an Uncharted movie.

Mark in this, for some strange reason I don't think this do well in the box office, but there is an chance it maybe successful. I would prefer Nathan Fillion as the hero since he has the same sense of humor as the character.
